Coating of wells with the specific binding biomolecule leaves unoccupied some sites on the bottom of wells. These sites must be blocked with specific step called “post-coating” in order to prevent non-specific binding of subsequent reactants. If this step is not performed, the assay will suffer from high background signal and lowered specificity and sensitivity.

Use of appropriate ELISA Blocking Solution is very important because, without the blocking step, the enzyme labeled secondary antibody can be adsorbed to the bottom of the plate in a non-specific manner causing a high background of the final reaction. On the contrary, by performing the blocking step the enzyme labeled secondary antibody binds only the antigen, creating a correct sandwich with the coated primary antibody. 

Biomat offer two types of blocking ELISA reagents:

  • ELISA Blocking: it’s a simultaneous microwell blocker and stabilizer buffer which works with both hydrophobic and hydrophilic ELISA plates. It preserves and protects biological activity of proteins and other biomolecules coated and dried on plate surface. Furthermore it prevents degradation, denaturation and leaching of proteins and other biomolecules. Biomat’s ELISA Blocking is a BSA (bovine serum albumin) solution containing a preservative and a stabilizing component.
  • BlockerWell: non proteic solution that maintains and protects the conformation and activity of dried proteins and other biomolecules coated on plate wells to prevent degradation, denaturation and leaching. Simultaneously blocks any free binding sites letf on the surface to reduce non-specific binding and therefore separate blocking of the surface is not necessary. 

Both Biomat’s ELISA Blocking Solutions are ready-to-use and efficiently block any free binding sites on the well surface after coating step.
